Caffeine: The Ingredient Everyone Already Knows

The FDA considers up to 400 mg per day not generally associated with dangerous negative effects in healthy adults. The European Food Safety Authority (EFSA) agrees, adding that single doses up to 200 mg raise no safety concerns for the general adult population. Most nootropic pouches contain somewhere between 50 and 150 mg of caffeine per serving, well within those ceilings when used as directed.

L-Theanine: The Calming Partner

L-theanine is an amino acid found naturally in tea leaves. Its job in a nootropic stack is to take the edge off caffeine's stimulant curve without blunting alertness. Human trials have used doses ranging from 100 to 400 mg with a consistently low side-effect profile. No serious adverse events have been attributed to L-theanine at supplemental doses in the published literature.

Methylliberine (Dynamine) and Theacrine (TeaCrine): What the Safety Trials Actually Found

These two purine alkaloids are structurally related to caffeine and show up together in several pouch formulas. The safety evidence is real, but thin, and the funding source matters.

Methylliberine

As of mid-2025, the PubMed record for methylliberine is approximately 9 papers, 5 involving humans. The largest safety trial (VanDusseldorp et al. 2020, Nutrients, n=125) tested 4 weeks of supplementation at 100-150 mg/day and found no clinically meaningful changes in heart rate, blood pressure, or blood chemistry markers. That is the highest dose and longest duration ever tested in people. Rat toxicology found a NOAEL of 150-225 mg/kg/day with no mutagenicity (Murbach 2019, PMID 31911801).

Theacrine

Theacrine has a slightly deeper evidence base. An 8-week trial (Taylor et al. 2016, Journal of the International Society of Sports Nutrition, n=60) at 200-300 mg/day found no habituation effect, meaning the compound did not lose effectiveness over time in the way caffeine typically does. But theacrine is not side-effect-free. At 6 mg/kg (roughly 400+ mg for an average adult), pooled human data showed increases in both systolic and diastolic blood pressure, along with GI discomfort, dizziness, and tremor (Lopes et al.). Rat toxicology established a NOAEL of 180 mg/kg/day, with deaths at 300-375 mg/kg/day from hepatocellular necrosis (Clewell et al. 2016).

The Funding Caveat

Most of the positive human research on both compounds was funded by Compound Solutions, the company that sells Dynamine and TeaCrine as branded ingredients. This does not automatically invalidate the findings, but it limits how much independent confirmation exists. Independently funded work on these alkaloids has been systematically less favorable. Read the data, but read the disclosures too.

Paraxanthine: The Newest Entrant

Paraxanthine is caffeine's primary metabolite. Your liver produces it every time you drink coffee. A handful of brands now use a synthesized version (branded as Enfinity) as a standalone stimulant, arguing it gives caffeine's benefits with fewer side effects.

The toxicology battery is clean. A toxicological battery (Purpura et al. 2021, Food Chem. Toxicol.; reviewed in Frontiers in Toxicology, 2023) found no mutagenicity, no genotoxicity, and no mortality or toxic effects in 28-day repeated-dose oral toxicity studies at up to 300 mg/kg bw/day in rats. Researchers reported that paraxanthine had a larger NOAEL than caffeine at equivalent testing doses, suggesting an improved safety margin relative to caffeine in animal models.

That said, paraxanthine's human trial history is shorter than caffeine's by decades. The animal data is encouraging. The long-term human data simply does not exist yet.

Alpha-GPC: The One Ingredient With an Open Safety Question

Alpha-GPC (L-alpha glycerylphosphorylcholine) is a choline donor used in some nootropic stacks to boost acetylcholine levels. It is the one ingredient on this list with a published safety signal that most product pages do not mention.

A 2021 study in JAMA Network Open analyzed the South Korean National Health Insurance Service database: 12,008,977 adults aged 50 and older, followed for 10 years. Alpha-GPC users showed a higher risk of total stroke (adjusted hazard ratio 1.43, 95% CI 1.41-1.46), ischemic stroke (aHR 1.34, 95% CI 1.31-1.37), and hemorrhagic stroke (aHR 1.37, 95% CI 1.29-1.46). The association was dose-dependent: more months of use correlated with higher risk.

The limitations are real and must be stated. This was an observational study, not a randomized trial. It cannot establish causation. The cohort was adults over 50, many of whom had pre-existing conditions. And the alpha-GPC was prescribed, often at clinical doses, not taken as a supplement by healthy younger adults. None of that erases the signal, but it puts boundaries around what the data actually shows.

Who Should Be More Cautious With Any Nootropic Pouch

Certain groups should talk to a clinician before using any stimulant-based pouch, regardless of the specific formula:

  • Pregnant or breastfeeding individuals. Caffeine crosses the placenta. The FDA and EFSA recommend pregnant people limit caffeine to 200 mg/day, and there is essentially zero pregnancy-specific data on methylliberine, theacrine, or paraxanthine.
  • People under 18. The FDA has not set a safe caffeine level for children or adolescents. No nootropic pouch has been studied in minors.
  • Anyone with a cardiovascular condition. Caffeine, theacrine, and paraxanthine all affect heart rate and blood pressure to varying degrees. If you have a diagnosed condition, ask your prescriber.
  • People on cognitive or cardiovascular medications. Stimulant interactions with MAOIs, blood thinners, or anti-hypertensives are not hypothetical. Talk to whoever writes your prescriptions.

This is not a list that any product label can override. A supplement brand telling you their formula is "safe for everyone" is a marketing claim, not a medical one.

The Dose Label Tells You More Than the Brand Name

Safety is not binary, and it is not settled by a brand's marketing page. The compounds in most nootropic pouches have real data behind them, ranging from decades of caffeine research to a handful of short-term trials for newer purine alkaloids. Alpha-GPC carries a published stroke-risk association that deserves plain disclosure, not burial. And every compound has a dose range where benefits flip to side effects.

The most useful thing you can do before buying any pouch is read the label. If a brand uses a proprietary blend and does not list exact milligrams per ingredient, you cannot evaluate safety at all. Transparency is the minimum bar.
